The orthodox Church of St. Spyridon in Peroj was built on the remains of the 16th century Church of St. Jerome, during a series of renovations that took place between 1788 and 1843. Its facade was redone in the 19th century classicist style while the interior was adapted to more resemble an Orthodox temple with the addition of an iconostasis (a rood screen) and icons of Greek saints. Upon completion the church was blessed and dedicated to St. Spyridon.

A 25 meter tall bell tower was further erected next to the church in 1860. While the Montenegrin settlers in Peroj visited the Church of St. Nicholas in Pula for their weekly liturgical activities, it took a hundred and thirty years for the community to get permission from the Venetian administration to build their own orthodox church in Peroj.

The Church of St. Spyridon is now a part of the Eparchy of Upper Karlovac.
